Diana Taurasi scored 21 of her game-high 23 points in the first 12 minutes and Sylvia Fowles had 18 overall.
Dominating the boards, pouring in points in transition and hitting shots from behind the arc, Spartak wasted no time in brushing aside Montpellier.
Taurasi scored seven of the team's first nine points as they opened up an early 9-2 lead and the two-time defending champions extended that to 17-6 midway through the first quarter.
With 1:20 to go in the initial frame, Taurasi took her points haul to 16 with another three to give the hosts a 31-11 advantage.
Spartak soon after reeled off the final six points of the quarter for a 37-14 lead.
Spartak eased up over the last several minutes of the first half and took a 56-21 lead to the break and cruised once play resumed.
Taurasi ended up five of six from long range and Spartak nailed 12 of 22 overall.
They out-rebounded Montpellier 42-24.
The loss dropped the French team to 1-9 and drew the curtain on their disappointing EuroLeague Women campaign.
Spartak will now shift their focus to the Eighth-Finals.
Diana Taurasi, Spartak Moscow Region player:
"We had to focus on this game because Montpellier is a better team than the results and statistics show. We are now happy and hoping that after the break we will continue playing well. I wish all a Merry Christmas and peace on earth."
Sheana Mosch, Lattes Montpellier player:
"Today our team could not compete with Spartak. I am disappointed we did not play better but … that is basketball. Merry Christmas and a Happy New Year."
László Rátgéber, Spartak Moscow Region coach:
"I am happy with my team as well as by the fact that we finished with the best record in the EuroLeague. Now we have to wait till the next year to continue the hard work. My warmest holiday wishes to all the world but especially our families and our fans."
Valéry Demory, Lattes Montpellier coach:
"Congratulations to Spartak and I wish them the best. I have seen most EuroLeague teams and without any doubt I can say Spartak is in a league of their own. As for the game my only comment is "touristic game". Our players enjoyed the trip and especially the Red Square."
